What is detected and measured
The KOMBITEST TBNK 6-colour reagent is intended for the detection and enumeration of lymphocyte populations and subsets in human whole blood by flow cytometry. It detects and measures relative percentages of human T cells (CD3+), B cells (CD3-CD19+), NK cells (CD3-CD16+56+), helper/inducer (CD3+CD4+) and suppressor/cytotoxic (CD3+CD8+) T cell subsets. It is intended for use in the immunological assessment of normal patients and might aid in the diagnosis of patients having, or suspected of having, an immune deficiency.
Absolute counts of the above subsets can be determined by one of two methods:
- Using the absolute lymphocyte count as determined by a haematology analyser, or
- Using the absolute volumetric count function of the XF-1600 flow cytometer.
The XF-1600 TBNK software on the XF-1600 flow cytometer provides automated lymphocyte subset population gating when used in combination with the KOMBITEST TBNK 6-colour reagent, and thus allows for single platform count analysis without the use of count beads.

The KOMBITEST TBNK 6-colour reagent is a ready-to-use premixed combination of the following fluorochrome-labelled murine monoclonal antibodies, diluted at optimum concentrations in a stabilising phosphate-buffered saline (PBS) solution containing 15 mM sodium azide:
